为获得毛葡萄遗传转化体系中合适的抗生素种类和体积质量分数,以毛葡萄单株“花溪-4”为试材,研究了选择抗生素潮霉素(Hygromycin,Hyg)、卡纳霉素(Kanamycin,Kan)和抑菌抗生素羧苄青霉素(Carbenicillin,Carb)及头孢霉素(Cefotaxime,Cef)对其离体叶片再生和再生芽成苗的影响.结果表明:0.1mg/L的Hyg和10mg/L的Kan可完全抑制不定芽再生,适合遗传转化中转基因再生芽筛选;再生芽在添加5mg/L Hyg和40mg/L Kan培养基中表现黄化致死,不能成苗,可用于转基因苗选择.300mg/L的Cef或400mg/L的Carb均可促进花溪-4不定芽再生,同时对根癌农杆菌抑制效果良好,适宜于毛葡萄遗传转化中抑制农杆菌生长.
In order to obtain the appropriate antibiotic species and volume fraction in the genetic transformation system of Gossypium hirsutum, the effects of selecting antibiotics hygromycin (Hyg), kanamycin (Kanamycin, Kan) and antibacterial antibacterials Carbenicillin (Carb) and cefotaxime (Cef) on in vitro leaf regeneration and regenerated bud formation were studied.The results showed that: 0.1mg / L Hyg and 10mg / L of Kan could completely inhibit the regeneration of adventitious buds, and was suitable for screening transgenic regenerated shoots in the transformation.
