目的 :采用子宫营养试验 ,检测双酚A(BisphenolA ,BPA)的雌激素样活性 ,初步探讨其作用机制。方法 :未成年SD雌性大鼠 (2 1d) 10 0只 ,分为BPA 2 0 0mg/kg、4 0 0mg/kg和 80 0mg/kg 3个剂量组 ,溶剂对照组 (花生油 )和阳性对照组 (苯甲酸雌二醇 ,EstradiolBenzoate,E2 B) 0 .4mg/kg ,每天一次灌胃给药 ,连续给予 3d和 7d。于末次给药的第 2 4h处死动物取子宫。结果 :3d和 7d两个时段 ,E2 B、BPA 4 0 0mg/kg和 80 0mg/kg 2个剂量组子宫湿重、子宫 /体重比、平滑肌厚度和宫腔上皮高度与溶剂对照组相比有显著性差异 (P <0 .0 1) ,有明显的剂量效应关系。给予BPA 7d还使未成年SD雌性大鼠阴道开口时间提前。结论 :BPA具有弱雌激素样效应 ,子宫湿重增加可能与平滑肌增生有关。
OBJECTIVE: To detect estrogen-like activity of bisphenol A (BPA) by uterine nutrition test and to explore its mechanism. Methods: A total of 100 female SD rats (21 days) were divided into 3 groups: BPA 200 mg / kg, 400 mg / kg and 80 0 mg / kg, and the solvent control group (peanut oil) and the positive control group (Estradiol benzoate, EstradiolBenzoate, E2 B) 0. 4mg / kg, once a day intragastric administration, continuous administration 3d and 7d. Animals were sacrificed at 24 hours after the last administration. Results: The uterine wet weight, uterine / body weight ratio, smooth muscle thickness and uterine epithelial height in two doses of E2 B, BPA 400 mg / kg and 80 0 mg / kg were significantly higher than those of the solvent control group at 3d and 7d Significant difference (P <0. 01), there is a clear dose-response relationship. Administration of BPA 7d also premature vaginal opening in minors SD female rats. Conclusion: BPA has a weak estrogen-like effect, and the increased uterine wet weight may be related to smooth muscle hyperplasia.
