建立了液相色谱-串联质谱法同时测定犬血浆中的替格瑞洛(1)及其主要活性代谢产物AR-C124910XX(2)。使用C18色谱柱,以乙腈∶2 mmol/L乙酸铵溶液(70∶30)为流动相,地西泮为内标,采用TQ-MS检测器,电喷雾离子源(ESI),以正离子多反应监测(MRM)方式进行定量分析,监测离子对m/z 523.3→m/z 127.1(1)、m/z 479.3→m/z 153.1(2)和m/z 284.9→m/z 193.0(内标)。1、2均在50~10 000 ng/ml范围内线性关系良好;提取回收率为109.8%~115.1%和109.3%~114.5%,日内RSD≤6.8%,日间RSD≤14.2%。
A liquid chromatography-tandem mass spectrometry (LC-MS / MS) method was developed for the simultaneous determination of ticagrelor (1) and its major active metabolite AR-C124910XX (2) in dog plasma. The mobile phase consisted of acetonitrile: 2 mmol / L ammonium acetate solution (70:30) and the internal standard was C18 column with TQ-MS detector and electrospray ionization source (ESI) (2) and m / z 284.9 → m / z 193.0 (m / z) were measured by MRM.
