目的 研究白介素 6 (IL 6 )在6 0 Coγ辐射诱导人多发性骨髓瘤细胞株XG 6、XG 7凋亡效应中的作用及其可能的分子生物学机理。方法 运用流式细胞仪技术 (FACS)、DNA片段释放法和RT PCR ,观察了IL 6对细胞株XG 6、XG 7中Bcl xL、BaxmRNA相对表达的影响及其与辐照诱导的细胞凋亡率的相关性。结果 IL 6存在时 ,XG 6、XG 7细胞的Bcl xL、BaxmRNA表达在辐照后迅速上升 ,4h达最高峰后逐渐下降 ,2 4~ 48h恢复到照射前水平 ;不加IL 6的XG 6、XG 7细胞的Bcl xL的表达在同样实验条件未检测到 ,而这 2株细胞BaxmRNA的表达在照射后不同时间均低于相应的加IL 6组 ,但尚有明显的表达。对不加IL 6的细胞组 ,辐照后不同时间的凋亡率明显高于相应的加IL 6组 (P <0 0 5 )。结论 IL 6可通过调节Bcl xL、Bax的表达及改变BcL xL Bax二者比率 ,保护辐照诱导XG 6、XG 7的细胞凋亡。
Objective To investigate the role of interleukin 6 (IL 6) in the apoptosis of human multiple myeloma cell lines XG 6 and XG 7 induced by 60 Co γ-rays and its possible molecular biological mechanism. Methods The effects of IL 6 on the relative expression of Bcl xL and Bax mRNA in XG 6 and XG 7 cell lines were observed by flow cytometry (FACS), DNA fragment release assay and RT PCR. Rate of relevance. Results In the presence of IL 6, the expression of Bcl xL and Bax mRNA in XG 6 and XG 7 cells increased rapidly after irradiation, reached the peak at 4 h and then decreased to the pre-irradiation level at 24 to 48 h. The expression of XG 6 The expression of Bcl xL in XG 7 cells was not detected under the same experimental conditions. However, the expression of Bax mRNA in these two cell lines was lower than that in the corresponding IL 6 group at different times after irradiation. The percentage of apoptosis at different time after irradiation was significantly higher than that of IL 6 group (P <0.05). Conclusion IL 6 can protect cells from XG 6 and XG 7 induced by radiation by regulating the expression of Bcl xL and Bax and the ratio of BcL xL Bax.
