莱氏野村菌产孢培养基的正交优化设计

研究不同碳源和氮源对莱氏野村菌[Nomuraea rileyi(Farlow)Samson]培养性状的影响,并筛选出产孢能力最强的培养基。以SMAY为基础培养基,分别更换碳源和氮源,研究培养性状的差异;采用正交试验对碳源和氮源含量进行优化。结果表明,不同碳源对莱氏野村菌有不同的作用,单糖类碳源有利于产孢,多糖类碳源有利于菌落生长,最有利于产孢的碳源是葡萄糖;酵母提取物是最有利于莱氏野村菌产孢的氮源,蛋白胨是最有利于菌落生长的氮源,蚕蛹粉与酵母提取物联合使用对产孢能力有增效作用。最有利于莱氏野村菌产孢的培养基组成为葡萄糖0.4%+酵母提取物1.6%+蚕蛹粉0.4%。 The effects of different carbon and nitrogen sources on the culture traits of Nomuraea rileyi (Farlow Samson) were studied, and the culture medium with the strongest sporulation ability was selected. Based on SMAY medium, carbon source and nitrogen source were changed respectively to study the differences of traits. The contents of carbon source and nitrogen source were optimized by orthogonal experiment. The results showed that different carbon sources had different effects on N. nivalis. The monosaccharide carbon source was favorable for sporulation. The polysaccharide carbon source was conducive to colony growth. The most favorable carbon source for sporulation was glucose. Yeast extract Is the nitrogen source most conducive to the spores production of Rhizoctonia solani. Peptone is the most suitable nitrogen source for colony growth. The combination of silkworm pupa powder and yeast extract has a synergistic effect on sporulation ability. The most conducive to the culture of Leishmania spore medium composed of 0.4% glucose + yeast extract 1.6% + silkworm pupa 0.4%.
