建立一种用高效液相色谱法(HPLC)测定茶叶中儿茶素类的方法,建立了同时分离测定了5种儿茶素组分(EGC、C、EGCG、EC、ECG)及咖啡碱(CAF)的方法,得到的最佳色谱条件为:ODS(5μm,4.6mm×250mm)色谱柱,流动相A:0.1%的磷酸甲醇溶液;流动相B:0.1%的磷酸水溶液;流速为1.0mL/min;柱温为30℃;UV检测波长278nm。儿茶素单体及咖啡碱在给定的浓度范围内线性关系良好,方法精密度和重复性的相对标准偏差在5%以内,测定样品中各标准品的平均加标回收率为98.20%—100.64%。与国标法GB/T 8313-2008相比,本方法解决了国标法中EC和EGCG难分离的问题。
A method for the determination of catechins in tea by high performance liquid chromatography (HPLC) was established. Five catechin components (EGC, C, EGCG, EC, ECG) and caffeine ). The optimal chromatographic conditions were as follows: ODS (5μm, 4.6mm × 250mm) column, mobile phase A: 0.1% phosphoric acid methanol solution; mobile phase B: 0.1% phosphoric acid aqueous solution; min; column temperature was 30 ℃; UV detection wavelength 278nm. Catechin monomer and caffeine in a given concentration range of a good linear relationship between precision and repeatability of the relative standard deviation of less than 5%, the determination of the standard samples of the average spike recovery was 98.20% -100.64 %. Compared with the national standard GB / T 8313-2008, this method solves the problem of difficult separation between EC and EGCG in GB method.
