【目的】基于数据挖掘技术筛选具有较明显抑制三阴性乳腺癌作用的中药。【方法】收集近15年关于乳腺癌中医治疗的有效文献,采用频次和聚类分析确定高频中药,制备其含药血清,采用四甲基偶氮唑盐(MTT)法和Transwell小室侵袭、迁移实验,分别观察各高频中药含药血清作用三阴性乳腺癌细胞(MDB-MB-231)12、24 h后细胞增殖、迁移和侵袭能力变化。【结果】筛选出的高频使用中药为川芎、蒲公英、白花蛇舌草、天花粉、牛膝。这5种中药的含药血清对MDB-MB-231细胞的增殖能力均具有不同程度的抑制作用,其中蒲公英、白花蛇舌草、牛膝对MDB-MB-231细胞迁移能力具有较强的抑制作用,蒲公英、川芎、白花蛇舌草对MDB-MB-231细胞侵袭能力具有明显抑制作用。【结论】运用数据挖掘技术得到的高频中药川芎、蒲公英、白花蛇舌草、天花粉、牛膝等具有明显的抑制三阴性乳腺癌作用。
【Objective】 The purpose of this study was to screen traditional Chinese medicine with obvious inhibitory effect on triple negative breast cancer based on data mining technology. 【Methods】 The effective literature about TCM treatment of breast cancer in recent 15 years was collected. The high frequency Chinese medicine was determined by frequency and cluster analysis, and the drug-containing serum was prepared by MTT assay and Transwell invasion. Migration assay was used to observe the changes of cell proliferation, migration and invasion after being treated with MDR-MD-231 cells for 12 and 24 hours respectively. 【Result】 The selected high-frequency Chinese herbs were Chuanxiong, Dandelion, Hedyotis diffusa, TCS and Achyranthes bidentata. The drug-containing serums of these five kinds of Chinese medicines have different degrees of inhibition on the proliferation of MDB-MB-231 cells, and the inhibitory effects of dandelion, Hedyotis diffusa and Achyranthes bidentata on the migration ability of MDB-MB-231 cells are stronger Effect of dandelion, Chuanxiong and Hedyotis diffusa on invasion of MDB-MB-231 cells was significantly inhibited. 【Conclusion】 Chuanxiong, dandelion, Hedyotis diffusa, TCS and Achyranthes bidentata, which were obtained by data mining technology, had significant inhibitory effect on triple negative breast cancer.
