目的:了解蓖麻毒蛋白(Ricin)及从大戟科植物蓖麻种子中提取出来的一种核糖体失活蛋白的热稳定性和抗肿瘤活性。方法:利用亲和层析的方法纯化蓖麻种子毒蛋白,并通过SDS-PAGE电泳和MTT染色法等方法检测其热稳定性、抗肿瘤性。结果:Ricin在37℃的缓冲液中保温60 h,没有明显的降解;能有效地杀死食管癌细胞EC109。结论:Ricin是有前途的蛋白类抗肿瘤药物。
Objective: To investigate the thermal stability and antitumor activity of Ricin and a ribosome inactivating protein extracted from Euphorbiaceae plant castor seeds. Methods: The toxic protein of castor seeds was purified by affinity chromatography and its thermal stability and antitumor activity were detected by SDS-PAGE electrophoresis and MTT staining. Results: Ricin incubated in buffer at 37 ℃ for 60 h showed no obvious degradation; it effectively killed esophageal cancer cells EC109. Conclusion: Ricin is a promising protein antitumor drug.
