目的和方法:研究肠道缺血再灌流过程对肾脏内源性碱性成纤维细胞生长因子(bFGF) 和转化生长因子β(bTGFβ) 基因表达的影响。采用肠系膜上动脉夹闭45 min 与再灌流6 h 和24 h 动物模型,用原位杂交与逆转录PCR(RT- PCR) 技术研究两种生长因子基因在正常、缺血以及再灌流肾组织中的表达。结果:两种因子的mRNA 在正常肾脏均有少量表达。缺血45 min 后,两种因子基因表达减少。再灌流6 h ,肾组织中TGFβ基因表达明显增加,而bFGF 基因表达仍然较少。再灌流24 h ,两种因子基因表达恢复至正常对照。结论:肠道缺血再灌流可以引起肾组织内源性bFGF 与TGFβ基因表达的改变,这种变化过程与肾缺血性损伤后自我修复机制密切相关
Objective: To investigate the effects of intestinal ischemia-reperfusion on the expression of basic fibroblast growth factor (bFGF) and transforming growth factor-β (bTGFβ) in kidneys. The animals were subjected to occlusion of the superior mesenteric artery for 45 min and reperfusion for 6 h and 24 h, respectively. In situ hybridization and reverse transcription-polymerase chain reaction (RT-PCR) were used to detect the expression of the two growth factor genes in normal, ischemic and reperfused renal tissues expression. Results: The mRNAs of the two factors were all expressed in small amount in normal kidneys. After 45 min of ischemia, the expression of the two factors decreased. At 6 h after reperfusion, the expression of TGFβ gene in renal tissue increased significantly, while the expression of bFGF gene was still low. After 24 h of reperfusion, the expression of two factor genes returned to the normal level. CONCLUSION: Intestinal ischemia-reperfusion can cause the changes of endogenous bFGF and TGFβ gene expression in renal tissues, which is closely related to the mechanism of self-repair after renal ischemic injury
