目的:研究滋阴地黄丸对兔眼脉络膜组织中血管内皮生长因子(VEGF),色素上皮衍生因子(bFGF),碱性成纤维细胞生长因子(PEDF)表达的影响。方法:用激光光凝建立兔眼脉络膜新生血管(CNV)动物模型,ig滋阴地黄丸9.75,39 g.kg-1,连续30 d后取眼球进行免疫组织化学染色,测定脉络膜中VEGF,bFGF及PEDF的表达。结果:模型对照组兔眼脉络膜组织中VEGF,bFGF呈强阳性表达及PEDF的弱阳性表达,滋阴地黄丸低、高剂量组中VEGF,bFGF的表达呈阳性或弱阳性,PEDF则呈阳性或强阳性表达。滋阴地黄丸低、高剂量兔眼脉络膜VEGF的表达为(1.20±0.45),(0.40±0.55)分,模型组为(2.60±0.55)分,(P<0.01);低、高剂量组bFGF的表达为(1.40±0.55),(0.40±0.55)分,模型组为(2.80±0.45)分,(P<0.01),低、高剂量组PEDF为(1.60±0.89),(2.80±0.45)分,模型组为(0.40±0.55)分(P<0.05,P<0.01)。滋阴地黄丸显著地抑制VEGF和bFGF的表达,诱导了PEDF的表达。结论:滋阴地黄丸通过抑制脉络膜中VEGF,bFGF的表达,诱导PEDF的表达而抑制了CNV的形成。
Objective: To study the effect of Ziyindihuang Pills on the expression of vascular endothelial growth factor (VEGF), bFGF and PEDF in the choroid of rabbits. Methods: Rabbit choroidal neovascularization (CNV) model was established by laser photocoagulation. Ig was injected with 9.75 and 39 g.kg-1 of Ziyindihuang Pills. After 30 days, the eyeballs were taken for immunohistochemical staining. The expressions of VEGF, bFGF And PEDF expression. Results: The expression of VEGF and bFGF in the choroidal tissues of the model control group was strongly positive and weakly positive in PEDF. The expression of VEGF and bFGF in the low and high doses of ZYRH group was positive or weakly positive while PEDF was positive or Strong positive expression. (1.20 ± 0.45) and (0.40 ± 0.55) in model group and (2.60 ± 0.55) in model group, respectively (P <0.01). In low and high dose group, the expression of bFGF (1.40 ± 0.55) and (2.40 ± 0.55) respectively in the model group and (2.80 ± 0.45) in the model group (P <0.01) Points, the model group was (0.40 ± 0.55) points (P <0.05, P <0.01). Ziyindihuang Pills significantly inhibited the expression of VEGF and bFGF and induced the expression of PEDF. Conclusion: Ziyindihuang pill can inhibit the formation of CNV by inhibiting the expression of VEGF and bFGF in choroid and inducing the expression of PEDF.
