目的:探讨骨髓细胞在受致死剂量照射的受体鼠体内的多向分化潜能。方法:将C57-TgN(Mx-cre)雄性小鼠的骨髓细胞经尾静脉植入受致死剂量照射的雌性受体鼠,重建受体鼠骨髓,然后分别用PCR和免疫组化分析检测受体鼠的多个器官组织中sry及Mx-cre基因的分布及表达情况。结果:PCR和免疫组化分析结果均显示,供体细胞的两个标志sry及Mx-cre在受体鼠的肝脏、肾脏、皮肤、脾脏等多个器官中都有表达。结论:骨髓细胞不仅可以重建受致死剂量照射受体鼠的造血系统,而且具有分化为肝脏、肾脏、皮肤、脾脏等组织细胞类型的多向潜能性。
Objective: To investigate the multidrug differentiation potential of bone marrow cells in recipient mice exposed to lethal doses. METHODS: Bone marrow cells of C57-TgN (Mx-cre) male mice were transplanted via the tail vein into the lethally irradiated female recipient mice to reconstitute the bone marrow of the recipient mice. PCR and immunohistochemistry were used to detect the receptor The distribution and expression of sry and Mx-cre genes in multiple organ tissues of mice. Results: The results of PCR and immunohistochemistry showed that the two markers of donor cells, sry and Mx-cre, were expressed in the liver, kidney, skin and spleen of recipient mice. CONCLUSION: Bone marrow cells can not only reconstruct the hematopoietic system of mice irradiated by lethal doses, but also have the multidirectional potential of differentiating into liver, kidney, skin and spleen cells.
