用多粘菌素B固定纤维治疗内毒素和感染性休克的新途径

内毒素在血液中循环是引起内毒素休克及其临床症象的重要因素。目前还没有一种药物能成功地治疗内毒素休克。木文旨在探讨使用新近研制的多粘菌素固定纤维(PMX-F)直接进行血液灌洗,能否有效地治疗狗内毒素和细菌性休克。将纤维截体和硫酸多粘菌素B混合或再加入一种肽缩合剂制成PMX-F。实验方法:1.PMX-F体外解毒能力的测定:在30 ml内毒素(ET)盐溶液内加入1gPMX-F使最终浓度达7.35、2、0.42和0.018 ng/ml,置37℃中90分钟。然后计算出ET残留浓度。2.PMX-F对鼠的作用:将2g PMX-F加入20mlLP5液(lipopolysac-charide)每ml水中含E·Coli 0111:B_40.5mg中, Endotoxin circulating in the blood is caused by endotoxic shock and its clinical symptoms as an important factor.
