重组IL-6和维甲酸诱导肾癌细胞凋亡的研究

目的 :探讨重组白细胞介素 6 (IL 6 )在体外对肾癌细胞GRC - 1(RCCGRC - 1)凋亡的诱导作用。方法 :在RCCGRC - 1细胞培养体系中加入IL 6诱导 ,反式维甲酸 (RA)作为对照药物 ,琼脂糖凝胶电泳和流式细胞仪观察IL 6和RA对细胞DNA电泳和细胞周期动力学的影响。结果 :IL 6诱发肾癌细胞DNA出现典型的梯状改变 ,RA有轻度诱导肾癌细胞凋亡作用。流式细胞仪DNA直方图上IL 6诱导肾癌细胞凋亡率明显高于RA(P <0 0 5 )。结论 :提示IL 6促进RCCGRC - 1凋亡的作用明显高于RA ,且随着剂量加大和时间增加 ,诱发凋亡程度增高 ,但需进一步探讨IL 6治疗肾癌的可能性及其价值 Objective: To investigate the induction of recombinant interleukin - 6 (IL - 6) on the apoptosis of renal cell carcinoma cell line GRC - 1 (RCCGRC - 1) in vitro. Methods: IL - 6 and trans - retinoic acid (RA) were added into the cell culture system of RCCGRC - 1 as controls, and the effect of IL - 6 and RA on DNA electrophoresis and cell cycle kinetics was observed by agarose gel electrophoresis and flow cytometry Impact. Results: The ladder-like changes of DNA in renal cell carcinoma induced by IL-6 were observed. RA had a slight effect on the apoptosis of renal cell carcinoma. Flow cytometry DNA histogram IL 6 -induced renal cell apoptosis rate was significantly higher than RA (P <0 05). Conclusion: It is suggested that the effect of IL - 6 on promoting the apoptosis of RCCGRC - 1 is obviously higher than that of RA. With the increase of dose and the increase of time, the degree of apoptosis induced is increased. However, the possibility and value of IL - 6 in treating renal cell carcinoma
