目的:建立同时测定三黄片(大黄、黄芩浸膏和盐酸小檗碱)中盐酸小檗碱和黄芩苷的含量测定方法。方法:采用高效液相色谱法,用C18色谱柱(150mm×4.6mm,5μm),以甲醇-0.05mol·L-1磷酸二氢钾溶液-冰乙酸(40:60:1)为流动相,流速为1.0ml·min-1,检测波长333nm。结果:盐酸小檗碱与黄芩苷的线性范围分别为0.34-1.68μg和1.0-5.0μg。平均加样回收率分别为98.31%和98.18%,RSD为0.98%和1.09%(n=6)。结论:本法操作简便,结果准确,分离效果好,可用于三黄片的质量控制。
Objective: To establish a method for the simultaneous determination of berberine hydrochloride and baicalin in Sanhuang tablets (rhubarb, Huangqi extract and berberine hydrochloride). Method: High performance liquid chromatography was used on a C18 column (150mm×4.6mm, 5μm) with methanol-0.05mol·L-1 potassium dihydrogen phosphate solution-glacial acetic acid (40:60:1) as the mobile phase. The flow rate was 1.0 ml·min-1 and the detection wavelength was 333 nm. Results: The linear ranges of berberine hydrochloride and baicalin were 0.34-1.68 μg and 1.0-5.0 μg, respectively. The average recovery rates were 98.31% and 98.18%, respectively, with RSDs of 0.98% and 1.09% (n=6). Conclusion: This method is simple, accurate, and has good separation effect. It can be used for quality control of Sanhuang tablets.
