目的:探讨表皮生长因子受体(EGFR)在隐睾组织中是否存在改变及可能发挥的作用。方法:妊娠SD大鼠随机分为3组,即正常对照组、玉米油对照组和邻苯二甲酸二乙基已基酯(DEHP)实验组,每组10只。玉米油溶剂对照组和DEHP实验组自妊娠第12.5日到妊娠第19.5日分别持续每日经口给予玉米油或DEHP(500 mg/kg)1 ml灌胃,正常对照组不作任何处理。收集出生后20 d的雄性仔鼠睾丸,采用免疫组织化学及Western blotting方法检测EGFR在睾丸组织中的表达。结果:正常对照组和玉米油对照组EGFR的表达无统计学差异(P>0.05);DEHP诱导隐睾组隐睾睾丸组织中EGFR表达与2个对照组相比明显增加,差异有统计学意义(P<0.05)。结论:孕期暴露环境内分泌干扰物DEHP可引起雄性子代发生隐睾,雄性隐睾子代睾丸组织中EGFR表达增加,可能与隐睾引起的远期不育有关。
Objective: To investigate whether epidermal growth factor receptor (EGFR) changes in cryptorchidism and its possible role. Methods: Pregnant SD rats were randomly divided into three groups: normal control group, corn oil control group and DEHP experimental group, with 10 rats in each group. The corn oil solvent control group and the DEHP experimental group were orally administered orally with corn oil or DEHP (500 mg / kg) 1 ml orally daily from the 12th day of gestation to the 19th day of gestation on the 19th day of gestation. The normal control group were given no treatment. The testis of male offspring was collected 20 days after birth and the expression of EGFR in testis was detected by immunohistochemistry and Western blotting. Results: The expression of EGFR in normal control group and corn oil control group had no significant difference (P> 0.05). The expression of EGFR in cryptorchid testis tissue of DEHP-induced cryptorchid group was significantly higher than that of the two control groups, the difference was statistically significant (P <0.05). Conclusion: Exposed to endocrine disruptor DEHP during pregnancy may cause cryptorchidism in male offspring. The expression of EGFR in male cryptorchid testis may be related to long-term infertility induced by cryptorchidism.
