应用聚合酶链反应(PCR)扩增幽门螺杆菌(HP)特异性的DNA片段,并与组织学、涂片法、尿素酶法同步分析和比较.结果显示HP的PCR扩增产物经琼脂凝胶电泳,均为468bp的单一区带,与原设计一致;与组织学、涂片法和尿素酶法的同步检测,发现PCR法的敏感性为94.4%,明显高于组织学60.9%、涂片法77.8%和尿素酶法86.6%.提示PCR法能快速、敏感、特异地检测临床标本中的HP,对探讨HP与上消化道疾病之间的关系、指导抗菌治疗均具有重要意义.
The specific DNA fragment of Helicobacter pylori (HP) was amplified by polymerase chain reaction (PCR), and analyzed by histology, smear method and urease method simultaneously.The results showed that the PCR product of HP was agar gel Gel electrophoresis were single band of 468bp, consistent with the original design; with histology, smear and urease simultaneous detection and found that the sensitivity of PCR was 94.4%, significantly higher than the histology of 60.9%, coated 77.8% of tablets and 86.6% of urease.It is suggested that PCR method can detect HP in clinical samples quickly, sensitively and specifically, and is of great significance in exploring the relationship between HP and upper gastrointestinal diseases and guiding antibacterial therapy.
