目的:探讨护肾固精方对慢性移植物肾病的影响。方法:以Fisher大鼠作供者,Lewis大鼠作受者进行同种肾移植。移植后治疗组大鼠隔日给予护肾固精方饲喂。移植后16周作肾功能、移植肾组织学检测,测定肾组织中转化生长因子mRNA的表达。结果:与对照组比较,移植后治疗组24 h尿蛋白持续保持较低的水平(P<0.01),血清肌酐水平较低(P<0.01),肌酐清除率高(P<0.01);移植肾肾小球硬化程度轻(P<0.01),Banff评分低(P<0.01);转化生长因子β1(TGF-β1)的mRNA表达减少(P<0.01)。结论:护肾固精方对移植肾功能有保护作用,其机制与影响TGFβ1在移植肾的表达有密切关系。
Objective: To investigate the effect of Hushen Gujing recipe on chronic allograft nephropathy. METHODS: All kidney transplantations were performed using Fisher rats as donors and Lewis rats as recipients. Rats in the treatment group were given a kidney-protecting Gujing formula on the next day. Renal function and histological examination of transplanted kidneys were performed 16 weeks after transplantation to determine the expression of transforming growth factor mRNA in renal tissue. Results: Compared with the control group, 24-hour urinary protein remained lower in the treated group (P<0.01), serum creatinine level was lower (P<0.01), and creatinine clearance was higher (P<0.01); Glomerular sclerosis was mild (P<0.01), Banff score was low (P<0.01), and the mRNA expression of transforming growth factor β1 (TGF-β1) was decreased (P<0.01). Conclusion: Hushen Gujing recipe has a protective effect on renal allograft function, and its mechanism is closely related to its effect on the expression of TGFβ1 in renal allografts.
