目的:采用星点设计-效应面法优化槐米中芦丁提取工艺。方法:以提取率及芦丁纯度为指标,以提取pH值、溶剂物料比及浸提时间为因素,设计星点实验;分别采用多元线性方程及二项式方程拟合实验结果,绘制三维效应面图及二维等高线图,确定因素最佳取值范围。结果:二指标与因素的关系均适合采用二项式方程拟合(r>0.9);按优化的条件操作,提取率为10.99%,芦丁纯度达100%,实验结果与模型预测值偏差小。结论:星点设计-效应面法用于优化芦丁提取工艺,模型预测的准确度高;所建立的提取工艺能耗低,产品纯度好,有较高的工业化应用价值。
OBJECTIVE: To optimize the extraction of rutin in Flos Sophorae by the combination of the star point design and response surface methodology. Methods: Taking the extraction rate and the purity of rutin as indexes, the star point experiment was designed by extracting the pH value, the ratio of solvent and material and the extraction time. The multivariate linear equation and binomial equation were used to fit the experimental results respectively and the three-dimensional effect Plan and two-dimensional contour map, determine the best value range. Results: The relationship between the two indexes and the factors was fit to fit with the binomial equation (r> 0.9). Under the optimized conditions, the extraction rate was 10.99% and the purity of rutin was 100%. The deviation of experimental results from the model predictive value was small . Conclusion: The star-point design-response surface method is used to optimize the extraction process of rutin, the prediction accuracy of the model is high. The established extraction process has low energy consumption, good product purity and high industrial application value.
