采用PCR-SSCP技术检测了39例膀胱癌及7例正常膀胱组织中Ha-ras基因、p53基因变异,并结合临床分期、病理分级等参数进行了综合分析。结果表明,膀胱癌组织中Ha-ras基因、p53基因变异率分别为36%、30.8%,两者均显著高于正常对照组(P<0.05);且与分级分期呈正相关。复发肿瘤的Ha-ras基因突变率显著高于原发肿瘤;6例标本同时呈现Ha-ras基因与p53基因突变。提示Ha-ras基因、p53基因变异及相互作用在很大程度上参与了膀胱癌的发生及演变,并与其生物学行为密切相关,有望成为判断肿瘤恶性程度、自然病程及预后的评估指标
PCR-SSCP was used to detect the mutations of Ha-ras gene and p53 gene in 39 cases of bladder cancer and 7 cases of normal bladder tissue. The combined analysis of clinical stage, pathological grading and other parameters were carried out. The results showed that the mutation rates of Ha-ras gene and p53 gene in bladder cancer tissues were 36% and 30.8%, respectively, which were significantly higher than those in normal control group (P <0.05). The mutation rate of Ha-ras gene in recurrent tumors was significantly higher than that in primary tumors. The mutations of Ha-ras gene and p53 gene were also found in 6 specimens. It is suggested that the mutation and interaction of Ha-ras gene and p53 gene are involved in the occurrence and evolution of bladder cancer to a large extent and are closely related to their biological behavior, which is expected to be an evaluation index to judge the degree of malignancy, natural course of disease and prognosis
