目的:考察硫酸铵梯度法制备马钱子碱脂质体的影响因素。方法:采用Sephadex G-50凝胶柱分离测定包封率,考察各工艺及处方因素对包封率的影响。结果:马钱子碱脂质体的包封率与硫酸铵浓度、乙醇体积、磷脂浓度等因素存在显著的正相关性。结论:硫酸铵梯度法可以用于马钱子碱脂质体制备并得到满意的包封率。硫酸铵浓度、乙醇体积和磷脂浓度是影响包封率的重要因素。
Objective: To investigate the factors affecting the preparation of brucine liposomes by ammonium sulfate gradient method. Methods: Sephadex G-50 gel column was used to determine the entrapment efficiency. The effects of various factors and formulation factors on entrapment efficiency were investigated. Results: The entrapment efficiency of brucine liposomes was positively correlated with the concentration of ammonium sulfate, ethanol volume, phospholipid concentration and other factors. Conclusion: The ammonium sulfate gradient method can be used for the preparation of strychnine liposomes with satisfactory encapsulation efficiency. Ammonium sulfate concentration, ethanol volume, and phospholipid concentration are important factors affecting encapsulation efficiency.
