目的:利用实时定量逆转录-聚合酶链反应(RT-PCR)技术来研究丹参酮Ⅰ对人胃癌MGC-803细胞中cmcy、p53、bcl-2、bax基因表达的影响,初步阐明丹参酮Ⅰ抑制胃癌细胞生长的作用机理。方法:将人胃癌细胞MGC-803接种于6孔板中培养24h,利用试剂盒提取细胞总RNA,通过逆转录方式获得稳定的DNA,采用RT-PCR法来检测丹参酮Ⅰ对细胞cmcy、p53、bcl-2、bax基因的差异性表达情况。结果:丹参酮Ⅰ显著下调了cmyc、bcl-2的基因表达,显著上调了p53、bax基因表达。结论:丹参酮Ⅰ通过下调cmyc、bcl-2以及上调p53bax基因表达,促进细胞凋亡,抑制人胃癌MGC-803肿瘤细胞的生长。
OBJECTIVE: To study the effect of tanshinone Ⅰ on the expression of cmcy, p53, bcl-2 and bax gene in human gastric cancer cell line MGC-803 by real-time reverse transcription-polymerase chain reaction The mechanism of cell growth. Methods: The human gastric cancer cell line MGC-803 was inoculated into a 6-well plate and cultured for 24 hours. Total RNA was extracted using a kit, and stable DNA was obtained by reverse transcription. The expressions of cmcy, p53, bcl-2, bax gene expression differences. Results: Tanshinone Ⅰ significantly down-regulated the gene expression of cmyc and bcl-2 and up-regulated the expression of p53 and bax gene. Conclusion: Tanshinone Ⅰ can inhibit the growth of human gastric cancer cell line MGC-803 by down-regulating cmyc, bcl-2 and up-regulating p53bax gene expression.
