目的建立测定保健食品中微胶囊型维生素A含量的反相高效液相色谱法。方法以适宜浓度氨水溶液破坏保健食品中微胶囊型维生素A的微胶囊结构,再以有机溶剂提取。采用Agilent ZORBAX SB-C18色谱柱(250 mm×4.6 mm,5μm),以甲醇为流动相,流速1.0 mL·min-1,检测波长为325 nm,柱温35℃。结果维生素A在1.41~56.4μg·mL-1内线性关系良好(r=1.000);平均回收率为97.5%(RSD=1.9%)。结论本法方法简单、准确,重现性好,适用于保健食品中微胶囊型维生素A的测定。
Objective To establish a reversed-phase high-performance liquid chromatographic method for the determination of the content of microencapsulated vitamin A in health food. Methods The microcapsule structure of microencapsulated vitamin A in health food was destroyed by suitable concentration of ammonia solution and then extracted with organic solvent. An Agilent ZORBAX SB-C18 column (250 mm × 4.6 mm, 5 μm) was used with methanol as the mobile phase at a flow rate of 1.0 mL · min-1 at a detection wavelength of 325 nm and a column temperature of 35 ° C. Results The linearity of vitamin A in the range of 1.41 ~ 56.4 μg · mL-1 was good (r = 1.000). The average recovery was 97.5% (RSD = 1.9%). Conclusion The method is simple, accurate and reproducible. It is suitable for the determination of microcapsules of vitamin A in health foods.
