目的 观察外源性视网膜母细胞瘤基因 (Rb基因 )对视网膜母细胞瘤 (retinoblastoma,RB)移植瘤细胞周期的影响。 方法 在建立裸鼠眼玻璃体腔 RB移植瘤模型及构建 Rb基因的逆转录病毒表达载体 PBabe- Rb的基础上 ,用脂质体 Dosper介导法将 Rb基因导入裸鼠 RB移植瘤 ,流式细胞仪检测 RB移植瘤细胞周期变化。 结果 Rb基因在 RB移植瘤内的表达使 RB移植瘤 G1 期细胞数明显增加 ,DNA指数及 S期百分比值有所降低。 结论 外源性 Rb基因可部分抑制 RB移植瘤细胞周期的进程。
Objective To observe the effects of exogenous retinoblastoma gene (Rb) on the cell cycle of retinoblastoma (RB) xenografts. Methods The model of RB xenografts in vitreous cavity of nude mice was constructed and the retroviral expression vector PBabe-Rb was constructed. The Rb gene was introduced into nude mice RB xenografts by flow cytometry The tumor cell cycle of RB tumor was examined. Results The expression of Rb gene in the xenografts of RB patients significantly increased the number of G1 phase cells and the percentage of DNA index and S phase in the tumor. Conclusion Exogenous Rb gene can partially inhibit the cell cycle progression of RB xenografts.
