目的探讨趋化因子CCL20及CCL22在狼疮鼠浆膜腔液中的表达及意义。方法以9只B6.MRL-Faslpr/J狼疮小鼠为观察组,6只同系C57BL/6J小鼠为对照组。取2组小鼠胸膜及腹膜腔液,用ELISA方法检测其中CCL20及CCL22的表达水平,并与小鼠血清中ANA、dsDNA抗体的含量以及小鼠24 h尿蛋白含量进行关联性分析。结果 CCL20在狼疮鼠浆膜腔液中高表达,与对照组相比两者差异有统计学意义(P<0.05),而CCL22与对照组相比两者差异无统计学意义(P>0.05);狼疮小鼠浆膜腔液CCL20含量与血清ds-DNA和24 h尿蛋白量呈正相关(P<0.05),与血清中ANA无明显相关性(P>0.05);而狼疮小鼠浆膜腔液CCL22含量与血清ds-DNA、ANA及24 h尿蛋白量均无明显相关性(P>0.05)。结论趋化因子CCL20可能参与狼疮鼠浆膜腔病变的发生,与狼疮鼠的病情活动、肾损害有一定相关性。
Objective To investigate the expression and significance of chemokines CCL20 and CCL22 in serous fluid of lupus. Methods Nine B6.MRL-Faslpr / J lupus mice were used as observation group and six C57BL / 6J mice as control group. The pleural and peritoneal fluid were collected from two groups. The expression levels of CCL20 and CCL22 were detected by ELISA, and the correlations were analyzed with the content of ANA and dsDNA in mouse serum and 24 h urinary protein in mice. Results CCL20 was highly expressed in serous mucosa of lupus erythematosus (P <0.05), while there was no significant difference between CCL22 and control group (P> 0.05). There was a positive correlation between serum CCL20 level and serum ds-DNA and 24 h urinary protein levels in lupus mice (P <0.05), but not serum ANA (P> 0.05); while lupus mouse serous cavity fluid CCL22 content and serum ds-DNA, ANA and 24 h urinary protein content were no significant correlation (P> 0.05). Conclusions The chemokine CCL20 may be involved in the pathogenesis of serosal lesions in lupus and has some correlation with the disease activity and renal damage in lupus mice.
