目的观察柴芪益肝颗粒(CQKL)对Hepa1-6肝癌细胞移植C57b l/6小鼠肝癌组织骨桥蛋白(OPN)及基质金属蛋白酶-2(MMP-2)表达的影响。方法构建C57b l/6小鼠肝癌模型,24只小鼠随机分为3组,观察组予柴芪益肝颗粒灌胃(273 mg/20 g),1次/d;空白对照组予生理盐水0.4 mL/20 g灌胃,1次/d;5-FU对照组采用腹腔注射5-FU[20 mg/(kg.d)];各组用药及处理均连续3周。观察各组抑瘤率、免疫组化法检测C57b l/6小鼠肝癌组织中OPN和MMP-2的表达。结果观察组和5-FU对照组的抑瘤率分别为38.52%和61.45%,与空白对照组比较,差异有统计学意义(P<0.01);观察组与5-FU对照组比较,差异有统计学意义(P<0.05),其作用弱于5-FU。柴芪益肝颗粒能抑制OPN和MMP-2的表达,与空白对照组比较差异有统计学意义(P<0.05)。结论柴芪益肝颗粒对肝移植瘤有一定的抑瘤及抗转移作用,其作用机制可能与肿瘤侵袭与转移密切相关的OPN和MMP-2表达有关,需要作进一步深入的研究。
Objective To observe the effect of Chai Yiyi Granule (CQKL) on the expression of osteopontin (OPN) and matrix metalloproteinase-2 (MMP-2) in hepatocellular carcinoma C57BL / 6 mice implanted with Hepa1-6 hepatoma cells. Methods C57BL / 6 mice model of hepatocellular carcinoma was established. Twenty-four mice were randomly divided into three groups. The observation group was treated with ChaiYiYiGan granules intragastrically (273 mg / 20 g once a day), while the blank control group was given normal saline / 20 g orally, once a day. The 5-FU control group was injected intraperitoneally with 5-FU [20 mg / (kg · d)] for 3 weeks. The inhibitory rate of tumor was observed in each group. The expression of OPN and MMP-2 in C57BL / 6 mice hepatocarcinoma was detected by immunohistochemistry. Results The inhibition rates of the observation group and the 5-FU control group were 38.52% and 61.45%, respectively, which were significantly different from those of the blank control group (P <0.01). There was significant difference between the observation group and the 5-FU control group Statistical significance (P <0.05), the effect is weaker than 5-FU. Chai Yiyi Gan particles can inhibit the expression of OPN and MMP-2, compared with the control group, the difference was statistically significant (P <0.05). Conclusions ChaiYiYiGan granules can inhibit the growth and metastasis of liver xenografts in a certain extent. The mechanism may be related to the expression of OPN and MMP-2, which are closely related to tumor invasion and metastasis, which needs further study.
