①目的为配合临床移植,将O'Brien法程序简化,以期达到同样的保存效果。②方法取16段猪新鲜主动脉,其中8段用O'Brien法,24h培养消毒后冷冻保存。冷冻保护剂为含有二甲基亚砜的M199液。8段采用改良法保存——即不进行24h培养消毒。新鲜标本、24h培养后及冷冻复温后分别取材行HE,Verhoef法染色行光镜及扫描电镜观察。③结果改良法深低温保存的主动脉壁大部分结构完整,与O'Brien法相比无明显差异。④结论组织学检查结果表明改良法保存的主动脉效果良好,与传统的方法相比简单、实用,可以在临床上推广使用
① The purpose of this project is to simplify O’Brien’s procedure with clinical transplantation so as to achieve the same preservation effect. Methods 16 pigs fresh aorta were taken, of which 8 were treated by O’Brien method, cultured for 24h, sterilized and stored frozen. Cryoprotectant is M199 solution containing dimethylsulfoxide. 8-segment improved method to save - that is not carried out 24h culture disinfection. Fresh specimens, 24h after culture and after freezing and rewarming were taken HE, Verhoef staining and scanning electron microscopy. Results improved method Cryopreservation of the aortic wall most of the structural integrity, compared with O’Brien no significant difference. ④ Conclusion The results of histological examination showed that the modified aortic preservation method is good, compared with the traditional method is simple and practical, it can be used clinically
