目的建立检测人胆汁中利奈唑胺浓度的HPLC法。方法以氯霉素为内标,色谱柱为Eclipse XDB C18(250 mm×4.6 mm,5μm),流动相为甲醇-水(45∶55),流速1.0 m L·min-1,柱温30℃,检测波长254 nm。结果利奈唑胺在质量浓度0.50~100 mg·L-1内线性关系良好(r=0.999 8),定量下限为0.50 mg·L-1;低、中、高浓度的日间、日内精密度RSD均<10%,相对回收率分别为98.62%、101.32%、100.64%,提取回收率>75%。结论本方法操作简便,结果准确,适用于胆汁中利奈唑胺的检测。
Objective To establish an HPLC method for the determination of linezolid in human bile. Methods Chloramphenicol was used as internal standard. The column was Eclipse XDB C18 (250 mm × 4.6 mm, 5 μm) with a mobile phase of methanol-water (45:55) at a flow rate of 1.0 mL · min-1 and a column temperature of 30 ℃ , Detection wavelength 254 nm. Results The linezolid had a good linearity (r = 0.999 8) with a lower limit of quantitation of 0.50 mg · L-1 at concentrations ranging from 0.50 to 100 mg · L-1. The intra-day and intra-day RSD The average recoveries were 98.62%, 101.32% and 100.64%, respectively. The recoveries were> 75%. Conclusion The method is simple, accurate and suitable for the determination of linezolid in bile.
