[目的]检测2型糖尿病胃轻瘫大鼠的血液代谢谱的变化,探讨代谢组学在糖尿病胃轻瘫研究中的应用。[方法]Wistar大鼠腹腔注射链脲菌素(Streptozotoein,STZ)80mg/kg 1周后,高糖高脂饲料喂养8周后,建立2型糖尿病胃轻瘫模型。动态检测空腹血糖(FBG)变化,三酰甘油(TG)、总胆固醇(TC)、游离脂肪酸(FFA)及胰岛素(INS)水平,采用气相色谱质谱联用(GC/MS)的方法检测造模前,造模1、2、4、6、8周后大鼠血液代谢谱的变化;并与正常对照组大鼠作比较。[结果]与正常对照组相比,模型组大鼠FBG持续升高,FFA、TG、TC明显升高,血清INS含量明显降低;模型组大鼠代谢谱与正常对照组完全区分,并随时间变化逐渐偏离;PLS-DA分析显示,2组的血液代谢物能很好的区别开。[结论]2型糖尿病胃轻瘫大鼠血液代谢组学发生明显变化,血液代谢组学在一定程度上反映其病理变化。
[Objective] To detect the changes of blood metabolites in type 2 diabetic gastroparesis rats and to explore the application of metabolomics in the study of diabetic gastroparesis. [Method] Wistar rats were intraperitoneally injected with Streptozotocin (STZ) 80mg / kg for 1 week. After fed with high glucose and high fat diet for 8 weeks, type 2 diabetic gastroparesis was established. The levels of fasting blood glucose (FBG), triglyceride (TG), total cholesterol (TC), free fatty acid (FFA) and insulin were detected dynamically by gas chromatography-mass spectrometry (GC / MS) Before and after 1, 2, 4, 6 and 8 weeks of modeling, the changes of blood metabolites in rats were compared with those in normal control rats. [Results] Compared with the normal control group, FBG of model rats kept increasing, FFA, TG, TC increased significantly, serum INS level decreased significantly. The metabolic profile of rats in the model group was completely distinguished from the normal control group, Changes gradually deviated; PLS-DA analysis showed that two groups of blood metabolites can be very good distinction. [Conclusion] The blood metabonomics changes obviously in type 2 diabetic gastroparesis, and blood metabonomics reflects its pathological changes to a certain extent.
