目的:探讨青蒿琥酯(Artesunate,Art)对大鼠肝星状细胞(HSC-T6)增殖和凋亡的影响。方法:选用活性良好的HSCT6细胞进行体外培养,应用倒置显微镜观察药物作用后的细胞形态改变;用CCK-8测定不同浓度、不同时间Art对该细胞增殖的抑制作用的影响;用AnnexinⅤ和PI双染的流式细胞术检测HSC-T6的细胞凋亡率的影响。结果:①倒置显微镜结果显示,Art作用过的贴壁细胞数目变少、突触变短变细、细胞表面变皱、由星形变成圆形。②12.5~200μg·ml~(-1)的Art在0~72 h范围内对HSC的增殖有明显的抑制作用(P<0.01),抑制率有时间和剂量依赖性。③流式细胞仪结果显示随着剂量、时间的增加,凋亡率显著增加(P<0.05)。结论:Art能抑制HSC-T6细胞的增殖,并可促进细胞凋亡。
Objective: To investigate the effect of Artesunate (Art) on the proliferation and apoptosis of rat hepatic stellate cells (HSC-T6). METHODS: HSCT6 cells were cultured in vitro. The morphological changes of cells were observed under inverted microscope. The effect of Art at different concentrations and time on the proliferation was determined by CCK-8. The effect of HSC-T6 on apoptosis was examined by flow cytometry. Results: (1) The results of inverted microscope showed that the number of Art adherent cells decreased, the number of adherent cells became shorter, the cell surface became wrinkled, and the star became circular. ② Art from 12.5 ~ 200μg · ml ~ (-1) inhibited the proliferation of HSC obviously in the range of 0 ~ 72 h (P <0.01), and the inhibition rate was time-and dose-dependent. ③Flow cytometry showed that with the dose and time increase, the apoptosis rate increased significantly (P <0.05). Conclusion: Art can inhibit the proliferation of HSC-T6 cells and promote apoptosis.
