目的本实验通过研究血小板源性生长因子(platelete-derived growth factor,PDGF)A,B,C,D在宫颈癌组织中的表达情况,分析其与宫颈癌临床分期和淋巴管新生的关系。方法采用实时荧光定量聚合酶链反应(real-time polymerase chain reaction,RT-PCR)法及免疫组化分析57例宫颈癌组织和20例正常宫颈组织中PDGF-A,B,C,D的表达情况及其与宫颈癌组织中的微淋巴管密度(lymphatic microvessel density,LMVD)的相关性。结果 PDGF-A,B,D在各期癌症组织中的表达水平均明显高于正常组织(P<0.05),而PDGF-C在正常组织与癌症组织中的表达无差异(P>0.05);PDGF-A在宫颈癌组织中表达随着临床分期的增加呈增高趋势,且具有统计学意义(P<0.05),余3个因子在这三组中的表达量均无统计学意义(P>0.05);正常宫颈组织中LMVD明显低于宫颈癌组织(P<0.05),且宫颈癌淋巴结转移组中的LMVD明显高于无转移组(P<0.05);宫颈癌组织中PDGF-A,B,D的表达与LMVD呈正相关性(P<0.05),但是三个因子在有淋巴结转移组与无转移组中的表达无差异(P>0.05)。结论 PDGF-A,B,D在宫颈癌的发生发展中起着重要的作用,同时PDGFs的表达也促进癌组织中淋巴管的新生,宫颈癌组织中淋巴管新生又是其发生淋巴结转移的早期阶段,因此通过阻断PDGFs的表达可能成为治疗宫颈癌的一个新靶点。
Objective To investigate the expression of platelet-derived growth factor (PDGF) A, B, C and D in cervical cancer and to analyze its relationship with clinical stage and lymphangiogenesis of cervical cancer. Methods The expressions of PDGF-A, B, C and D in 57 cases of cervical cancer and 20 cases of normal cervical tissue were analyzed by real-time polymerase chain reaction (RT-PCR) and immunohistochemistry And its relationship with lymphatic microvessel density (LMVD) in cervical cancer tissues. Results The expression of PDGF - A, B and D in each stage of cancer was significantly higher than that in normal tissues (P <0.05), while there was no difference in the expression of PDGF - C between normal tissues and cancerous tissues (P> 0.05). The expression of PDGF-A in cervical cancer tissues increased with the increase of clinical stage, and had statistical significance (P <0.05). The expression of PDGF-A in these three groups had no statistical significance (P> 0.05). The LMVD in normal cervical tissue was significantly lower than that in cervical cancer (P <0.05), and the LMVD in cervical lymph node metastasis was significantly higher than that in non-metastatic cervical cancer (P <0.05) , While the expression of D was positively correlated with LMVD (P <0.05). However, there was no difference in the expression of three factors between lymph node metastasis and non-metastasis (P> 0.05). Conclusions PDGF-A, B and D play an important role in carcinogenesis and progression of cervical cancer. At the same time, the expression of PDGFs promotes the lymphangiogenesis in cervical cancer. Lymphangiogenesis in cervical cancer is the early stage of lymph node metastasis Stage, thus blocking the expression of PDGFs may become a new target for the treatment of cervical cancer.
