转染P21~(WAF1)基因对GRC—1肾癌细胞凋亡的影响

目的:建立GRC—1肾癌细胞凋亡模型并探讨P21WAF1基因对肾癌细胞凋亡的影响。方法:采用顺铂诱导法建立肾癌细胞凋亡模型及转染P21WAF1基因方法观察P21WAF1基因的作用。结果:①用20μmol顺铂诱导GRC—1肾癌细胞。建立了细胞凋亡模型,凋亡细胞占细胞总数为29.4%。②成功转染了P21WAF1基因至GRC—1肾癌细胞;③转染P21WAF1基因的GRC—1肾癌细胞,培养至第10代细胞加入20μmol顺铂后,凋亡细胞占细胞总数为75.6%。④凋亡细胞比率29.4%与75.6%之间存在显著性差异,P<0.01。结论:①成功建立了肾癌细胞凋亡模型;②转染P21WAF1基因对肾癌细胞凋亡有促进作用。 Objective: To establish a model of renal cell carcinoma of GRC-1 and to investigate the effect of P21WAF1 gene on apoptosis of renal cell carcinoma. Methods: Apoptosis model of renal cell carcinoma was established by cisplatin induction and the effect of P21WAF1 gene was observed by transfecting P21WAF1 gene. Results: ① GRC-1 cells were induced by 20μmol of cisplatin. Established a model of apoptosis, the total number of apoptotic cells accounted for 29.4% of cells. ②Transfection of P21WAF1 gene into GRC-1 cells; ③Transfection of GRP-1 cells with P21WAF1 gene into the10th generation cells, after adding20μmol / L cisplatin, the number of apoptotic cells accounted for75.6%. ④ There was a significant difference between apoptotic cells ratio of 29.4% and 75.6%, P <0.01. Conclusion: ① The model of renal cell carcinoma has been successfully established; ② The transfection of P21WAF1 gene can promote the apoptosis of renal cell carcinoma.
