目的:探讨苦瓜总皂苷对2型糖尿病大鼠心肌组织中血红素加氧酶1(Heme Oxygenase1,HO-1)和基质金属蛋白酶-9(Matrix Metallo Protein-9,MMP-9)表达的影响。方法:将60只健康雄性SD大鼠,随机分为正常组、模型组以及苦瓜总皂苷治疗组,模型组和苦瓜总皂苷治疗组给予高脂饮食4周,腹腔一次性注射STZ 30 mg建立2型糖尿病大鼠模型,造模成功后,治疗组给予苦瓜总皂苷(20 mg/kg)灌胃治疗,其它两组给予生理盐水灌胃,分别于治疗4周和8周后处死大鼠各半,检测血糖、心功能以及心肌组织HO-1和MMP-9的表达。结果:与正常对照组相比,模型组血糖,心功能CK、CK-MB、LDH和HBDH明显升高(P<0.05),治疗4周后均明显降低(P<0.05),治疗8周后进一步降低。心肌组织HO-1和MMP-9免疫组化、Western以及RT-PCR可见,与正常组相比糖尿病组HO-1表达明显下降(P<0.05),MMP-9明显升高(P<0.05);治疗后HO-1表达明显增加(P<0.05),MMP-9表达明显减少(P<0.05)。结论:苦瓜总皂苷对大鼠心肌组织中MMP-9和HO-1有明显的调控作用。
Objective: To investigate the effect of bitter gourd saponins on the expression of heme oxygenase 1 (HO-1) and matrix metalloproteinase-9 (MMP-9) in myocardium of type 2 diabetic rats. Methods: Sixty healthy male Sprague-Dawley rats were randomly divided into normal group, model group and total saponin of Momordica charantia. The model group and the total saponin of Momordica charantia were given high-fat diet for 4 weeks and intraperitoneal injection of STZ 30 mg After the model was established successfully, the treatment group was given gavage (20 mg / kg) gavage, and the other two groups were given normal saline gavage. The rats were sacrificed at 4 and 8 weeks after treatment, respectively The blood glucose, heart function and the expression of HO-1 and MMP-9 in myocardium were detected. Results: Compared with the normal control group, the levels of CK, CK-MB, LDH and HBDH were significantly increased (P <0.05) in the model group and significantly lower after 4 weeks (P <0.05) . The expression of HO-1 and MMP-9 in myocardium were detected by Western blotting and RT-PCR. The expression of HO-1 in diabetic group was significantly lower than that in normal group (P <0.05) ; After treatment, the expression of HO-1 increased significantly (P <0.05), and the expression of MMP-9 decreased significantly (P <0.05). Conclusion: Total saponin of Momordica charantia possesses obvious regulation on MMP-9 and HO-1 in myocardial tissue of rats.
