目的合成Keggin型硅钨酸盐K_4[α-SiW_(12)O_(40)]·17H_2O(简称KSiW_(12)),研究其体外抗肿瘤活性。方法通过IR、UV等表征手段对其结构进行表征,并应用四甲基偶氮唑盐(3-[4,5-dimethylthiazo-2-y]-2,5-diphenyltetrazolium bromide,MTT)比色法分析KSiW_(12)对人肝癌细胞(HepG2)以及人肺癌细胞(A549)的增殖抑制活性。采用光学显微镜观察两种肿瘤细胞在不同浓度药物作用后的形态变化。结果 Keggin型硅钨酸盐KSiW_(12)对两种肿瘤细胞均有增殖抑制作用,具有药物浓度依赖性,对两种肿瘤细胞的IC_(50)值分别为(7.5×10~(-5)±3.2)mol/L和(1.7×10~(-4)±3.8)mol/L。光学显微镜下经药物作用后的肿瘤细胞出现皱缩、变圆、缩小等形态变化。结论硅钨酸盐KSiW_(12)具有体外抗肿瘤活性,能够抑制肿瘤细胞增殖。
OBJECTIVE To synthesize Keggin-type silicotungstate K_4 [α-SiW_ (12) O_ (40)] · 17H_2O (KSiW_ (12) for its in vitro antitumor activity. Methods The structures were characterized by means of IR and UV. The structures of the compounds were characterized by MTT colorimetry The inhibitory activity of KSiW_ (12) on human hepatocellular carcinoma cells (HepG2) and human lung cancer cells (A549) was analyzed. The morphological changes of two kinds of tumor cells after different concentrations of drugs were observed by light microscope. Results Keggin type silicate tungstate KSiW 12 inhibited the proliferation of both tumor cells in a concentration-dependent manner. The IC 50 values of the two tumor cells were (7.5 × 10 -5) ± 3.2) mol / L and (1.7 × 10 ~ (-4) ± 3.8) mol / L. Under the light microscope, the tumor cells after shrinkage, rounding, shrinking and other morphological changes were observed. Conclusion The silicotungstate KSiW_ (12) has anti-tumor activity in vitro and can inhibit tumor cell proliferation.
