目的通过检测缺血性性脑卒中患者与正常对照组患者外周血中mi RNA125a-5p的表达水平,探讨其与与疾病发生的相关性及作为分子标记的可能性。方法该病例对照研究纳入30例有缺血性脑卒中的病例受试者以及26例对照受试者。收集外周血,提取总mi RNA,用QRT-PCR法检测mi R125a-5p表达水平。比较两组外周血中mi R125a-5p表达水平的差异。进一步按照病程、严重程度及OCSP亚型分组进行分析比较。结果脑梗死患者外周血中的mi R125a-5p表达水平较正常对照组外周血的mi R125a-5p明显下降(P<0.001);4个病程中,急性期组mi R125a-5p的表达水平显著降低(P<0.05);从严重程度分析,轻型组中mi R125a-5p的表达水平显著降低(P<0.05);根据OCSP亚型分组分析比较,TACI组的mi R125a-5p明显升高,差异有统计学意义(P<0.05);PACI组和LACI组mi R125a-5p的表达水平显著降低,差异有统计学意义(P<0.05)。结论 mi R125a-5p在缺血性脑卒中患者外周血中表达异常,明显下降,提示mi R125a-5p可能与缺血性脑卒中相关。
Objective To detect the expression of mi RNA125a-5p in the peripheral blood of patients with ischemic stroke and normal controls, and to explore its relationship with the disease and the possibility of using it as a molecular marker. Methods This case-control study included 30 patients with ischemic stroke and 26 control subjects. Peripheral blood was collected and total miRNA was extracted. The expression of mi R125a-5p was detected by QRT-PCR. The differences of mi R125a-5p expression in peripheral blood between the two groups were compared. Further according to the course of disease, severity and OCSP subgroup analysis and comparison. Results The expression of mi R125a-5p in the peripheral blood of patients with cerebral infarction was significantly lower than that of the normal control group (P <0.001). The expression of mi R125a-5p in the acute phase group was significantly lower than that of the normal control group (P <0.05). From the severity analysis, the expression level of mi R125a-5p in the light group was significantly decreased (P <0.05). According to OCSP subtype analysis, the mi R125a-5p in the TACI group was significantly increased (P <0.05). The expression of mi R125a-5p in PACI group and LACI group was significantly lower than that in control group (P <0.05). Conclusion The expression of mi R125a-5p in peripheral blood of patients with ischemic stroke is abnormal and obviously decreased, which suggests that mi R125a-5p may be associated with ischemic stroke.
