RNA干涉Survivin基因对SiHa细胞增殖抑制及凋亡的影响

目的:研究RNA干涉survivin基因对宫颈癌SiHa细胞的增殖抑制及凋亡的影响并探讨其作用机制。方法:重组质粒(pU6-siRNA-Survivin)及空质粒转染SiHa细胞,MTT法检测细胞增殖情况,检测survivin基因转录水平,吖啶噔染色、流式细胞术检测细胞凋亡,免疫组化染色检测survivin、caspase3基因表达。结果:RNA干涉后,抑制SiHa细胞增殖并出现细胞凋亡,survivin基因转录表达下调,caspase3基因表达上调。结论:RNA干涉survivin基因可导致SiHa细胞增殖抑制并凋亡,其机制与caspase3基因表达上调有关。 Objective: To study the effect of RNA interference survivin gene on the proliferation and apoptosis of cervical cancer SiHa cells and to explore its mechanism. Methods: The recombinant plasmids (pU6-siRNA-Survivin) and empty plasmid were transfected into SiHa cells. The proliferation of SiHa cells was detected by MTT assay. The transcription of survivin gene was detected by flow cytometry. Apoptosis was detected by flow cytometry. Detection of survivin, caspase3 gene expression. Results: After RNA interference, SiHa cell proliferation was inhibited and apoptosis occurred. Survivin gene transcription was down-regulated and caspase3 gene expression was up-regulated. Conclusion: RNA interference survivin gene can cause SiHa cell proliferation inhibition and apoptosis, the mechanism of which is related to the up-regulation of caspase3 gene expression.
