目的:观察单味中药泰山白花丹参提取物(SBE)对人脐血内皮祖细胞(EPCs)增殖、黏附及分泌一氧化氮(NO)的影响。方法:采用密度梯度离心法从脐血获得单个核细胞,将其接种在人纤维连接蛋白包被的培养板上,培养7d后收集贴壁细胞,加入不同浓度SBE继续培养24h;分别采用MTT比色法、黏附能力测定实验和硝酸还原酶法测定EPCs的增殖、黏附和分泌NO能力。结果:与对照组比较,SBE组EPCs增殖、黏附能力提高,培养上清中NO浓度增加(P<0.05),其作用在一定范围内随SBE浓度增加而增强。结论:白花丹参显著改善EPCs功能,这可能是其内皮保护的新机制。
OBJECTIVE: To observe the effect of single Chinese medicinal herb Taishan white salvia miltiorrhiza (SBE) on proliferation, adhesion and secretion of nitric oxide (NO) in human umbilical cord blood endothelial progenitor cells (EPCs). Methods: Mononuclear cells were obtained from umbilical cord blood by density gradient centrifugation and inoculated on human fibronectin-coated plates. After 7 days of culture, adherent cells were collected and incubated with different concentrations of SBE for 24 hours. MTT ratios Colorimetric assay, adhesion assay and nitric acid reductase assay for EPCs proliferation, adhesion and secretion of NO ability. Results: Compared with the control group, the proliferation and adhesion of EPCs in SBE group increased and the concentration of NO in culture supernatant increased (P <0.05). The effect of SBE increased with the increase of SBE concentration in a certain range. Conclusion: Salvia miltiorrhiza markedly improves EPCs function, which may be a new mechanism of endothelial protection.
