黄芪多糖对糖尿病足部溃疡成纤维细胞增殖功能的影响

目的探讨黄芪多糖(APS)能否恢复糖尿病足部溃疡成纤维细胞(Fib)的增殖功能。方法以糖尿病急性伤口为对照,在了解糖尿病足部溃疡伤口炎症因子白介素1β(IL-1β)变化的基础上,进而探讨APS对溃疡处IL-1β含量的变化及Fib增殖功能的影响。结果 (1)糖尿病足溃疡患者创面渗出液中IL-1β含量为(89.16±38.14)ng/ml,糖尿病急性损伤患者创面渗出液中IL-1β含量为(8.84±7.06)ng/ml,两者比较,差异有统计学意义(P<0.01)。予黄芪多糖500μg/ml外敷足部溃疡14 d后IL-1β降至(11.67±3.12)ng/ml,与治疗前相比有显著差异(P<0.01),而与急性伤口相比却差异不显著(P>0.05)。(2)IL-1β(0.5~5 ng/ml)浓度范围可明显促进Fib增殖,随着浓度增加,50 ng/ml IL-1β对Fib增殖没有影响(P>0.05).IL-1β增加到500 ng/ml则对Fib增殖抑制作用(3)APS100μg/ml(vs IL-1β 50ng/ml组,P=0.003 vs 对照组P=0.002)和APS500 μg/ml(vs IL-1β 50 ng/ml组P=0.000,vs对照组P=0.000)可明显促进Fib增殖。结论 IL-1β在低浓度(0.5~5 ng/ml)对成纤维细胞的增殖有促进作用,当浓度增到50 ng/ml时对细胞增殖没有刺激作用。随着浓度进一步升高(500 ng/ml)时对细胞增殖有明显抑制作用。APS在100μg/ml以上浓度可显著刺激糖尿病足部溃疡部位成纤维细胞的增殖。这种作用有可能是通过降低渗出液中IL-1β浓度实现的。 Objective To investigate whether Astragalus polysaccharide (APS) can restore the proliferation of diabetic foot ulcers fibroblasts (Fib). Methods Based on the changes of interleukin-1β (IL-1β) in wounds of diabetic foot ulcers, the changes of IL-1β in ulcer and the proliferation of Fib were studied. Results (1) The content of IL-1β in wound exudate of diabetic foot ulcer patients was (89.16 ± 38.14) ng / ml, while the level of IL-1β in wound exudate of diabetic patients with acute injury was (8.84 ± 7.06) ng / The difference between the two groups was statistically significant (P <0.01). The level of IL-1β was decreased to (11.67 ± 3.12) ng / ml after APS 500μg / ml for 14 days in APS group (P <0.01), but not significantly different from that in acute wounds Significant (P> 0.05). (2) IL-1β (0.5-5 ng / ml) could significantly promote the proliferation of fibroblasts.With the increase of concentration, 50 ng / ml IL-1βhad no effect on Fib proliferation (P> 0.05) 500 ng / ml inhibited proliferation of Fib. (3) APS100μg / ml (vs IL-1β 50ng / ml, P = 0.003 vs control P = 0.002) and APS500μg / ml Group P = 0.000, vs control group P = 0.000) could significantly promote the proliferation of Fib. CONCLUSION: IL-1β can promote the proliferation of fibroblasts at low concentration (0.5-5 ng / ml). When the concentration of IL-1β is increased to 50 ng / ml, the proliferation of fibroblasts is not stimulated. With further increase of concentration (500 ng / ml), the cell proliferation was significantly inhibited. APS at a concentration above 100μg / ml can significantly stimulate the proliferation of diabetic foot ulcer fibroblasts. This effect is likely to be achieved by reducing the concentration of IL-1β in the exudate.
