目的分析中国炭疽芽胞杆菌致死因子基因中的单核苷酸多态性(SNP)位点特征。方法选择18株炭疽芽胞杆菌,采用PCR方法扩增炭疽芽胞杆菌致死因子基因、测序,并进行SNP位点分析。结果实验中所用中国炭疽芽胞杆菌致死因子基因中第895位核苷酸均为G,与国外菌株不同。中国不同地区(包括新疆、广西、北京、内蒙古等地区)、不同年代和不同来源(包括土壤、牛、羊、骡、狗、患者等)分离的菌株,具有相同的SNP位点特征。结论致死因子基因中第895位核苷酸均为G的SNP位点特征很可能是中国炭疽芽胞杆菌的共同特征。
Objective To analyze single nucleotide polymorphism (SNP) loci in lethal factor genes of Bacillus anthracis in China. Methods 18 strains of Bacillus anthracis were selected and the lethal gene of Bacillus anthracis was amplified by PCR and sequenced. The SNP loci were also analyzed. Results The 895th nucleotide of the lethal factor gene of Bacillus anthracis in China was all G, which was different from that of foreign strains. Strains isolated in different regions of China (including Xinjiang, Guangxi, Beijing, Inner Mongolia, etc.), from different generations and from different sources (including soil, cattle, sheep, mules, dogs, patients, etc.) share the same SNP site characteristics. Conclusion The SNP locus of G at nucleotide 895 in lethal factor gene is likely to be a common feature of B. anthracis in China.
