以响叶杨×84K杨的茎段和叶片为外植体,应用正交实验方法对响叶杨×84K杨的再生条件进行研究。结果表明:茎段和叶片不定芽诱导的最佳培养基分别为MS+TDZ 0.0015mg/L+NAA0.05 mg/L,茎段不定芽再生率可达91%,MS+TDZ 0.001 mg/L+NAA0.01mg/L,叶片不定芽再生率可达70%;诱导不定芽生根的最佳组合为1/2MS+NAA0.01 mg/L+IBA0.1 mg/L,生根率可达100%。在碳源对生根率影响的研究中,蔗糖的影响最大,生根率可达88.9%。
The stem segments and leaves of Populus × eucalyptus × 84K were used as explants. The regeneration conditions of Populus × euphratica × 84K poplar were studied by orthogonal experiment. The results showed that the optimal medium for adventitious bud induction was stem segment and leaf explant, respectively. MS + TDZ 0.0015mg / L + NAA 0.05mg / L, regeneration rate of stem adventitious buds up to 91%, MS + TDZ 0.001 mg / L + NAA0.01mg / L, the regeneration rate of adventitious buds was up to 70%. The optimal combination of adventitious buds rooting was 1 / 2MS + NAA0.01 mg / L + IBA0.1 mg / L, the rooting rate could reach 100% . In the study of the effect of carbon source on rooting rate, sucrose had the greatest impact, with the rooting rate reaching 88.9%.
