目的 探讨诱导肾癌细胞凋亡的新方法及其机制。方法 采用HE染色、荧光染色、琼脂糖电泳等方法 ,分别对Cryptophycin(CPP)不同的药物浓度 ( 0 ,1,5 ,10 ,5 0 pmol/L)和不同培养时间段 ( 4~ 2 4h)所诱导的凋亡细胞形态、出现时间和密度比例进行观察和比较。结果 在光镜下能观察到CPP诱导的肾癌细胞凋亡的典型形态 ,用AO/EB荧光染色能区分出 4种不同细胞 ,DNA电泳证实有DNA裂解的现象。CPP在 10 pmol/L水平诱导细胞凋亡比率较高 ,作用 12h已基本达到凋亡的较高水平。结论 CPP可诱导肾癌细胞凋亡的发生 ,药效敏感。
Objective To explore a new method of inducing apoptosis of renal cell carcinoma and its mechanism. Methods The effects of Cryptophycin (CPP) at different concentrations (0, 1, 5, 10, 50 pmol / L) and different incubation periods (4 ~ 24 h) were studied by HE staining, fluorescent staining and agarose gel electrophoresis. The induced apoptotic cell morphology, appearance time and density ratio were observed and compared. Results Under light microscopy, CPP-induced apoptosis of renal cell carcinoma was observed. Four kinds of cells were identified by AO / EB staining, and DNA fragmentation was confirmed by DNA electrophoresis. CPP induced a higher apoptosis rate at 10 pmol / L, and reached a higher level of apoptosis at 12 h. Conclusion CPP can induce the apoptosis of renal cell carcinoma, and is sensitive to the effect.
