目的:研究ArgGlyAspSer(RGDS)肽对血小板聚集和血小板膜GPⅡb/Ⅲa反磷酸化的影响。方法:免疫沉淀提取血小板膜GPⅡb/Ⅲa,反磷酸化测定。结果:50μmol·L-1ADP引起血小板聚集时,其GPⅡb/Ⅲa反磷酸化明显降低。而应用100和200μmol·L-1RGDS肽与ADP共同孵育,呈浓度依赖抑制血小板聚集的同时,可逆转ADP引起的蛋白反磷酸化改变(P<0.01),其抑制血小板聚集和GPⅡb/Ⅲa反磷酸化呈明显的负相关(r=-0.76,P<0.01)。结论:RGDS肽抑制血小板聚集是通过抑制血小板膜GPⅡb/Ⅲa磷酸化所致
Objective: To study the effect of ArgGlyAspSer (RGDS) peptide on platelet aggregation and GPⅡb / Ⅲa phosphorylation. Methods: Platelet membrane GPⅡb / Ⅲa was extracted by immunoprecipitation, and its anti-phosphorylation was measured. Results: When platelet aggregation was induced by 50μmol·L-1ADP, the anti-phosphorylation of GPⅡb / Ⅲa was significantly decreased. However, incubation of 100 and 200 μmol·L-1 RGDDS peptides with ADP inhibited the aggregation of platelets in a concentration-dependent manner and reversed the phosphorylation of protein induced by ADP (P <0.01), which inhibited platelet aggregation and GPⅡb / Ⅲa Anti-phosphorylation showed a significant negative correlation (r = -0.76, P <0.01). Conclusion: RGDS peptide inhibits platelet aggregation by inhibiting platelet membrane GP Ⅱ b / Ⅲ a phosphorylation
