目的:探讨生骨紫金丹的作用机理,为其进一步临床应用提供依据。方法:将W istar大鼠72只造骨折模型后随机分为模型空白组,生骨紫金丹组,阳性对照组。于造模后第2周、3周、4周取局部骨痂制成石蜡切片,以免疫组化技术显示TGF-β1和bFGF,进行图象定量分析。结果:在骨折愈合过程,骨痂中可见TGF-β1和bFGF的阳性表达,灰度测定结果显示在第2周、3周TGF-β1和bFGF的阳性表达强度生骨紫金丹组和阳性对照组与模型空白组有高度显著性差异(P<0.01)。结论:生骨紫金丹能促进TGF-β1和bFGF的表达,促进骨折愈合。
Objective: To explore the mechanism of the action of Shenggu Zijindan and provide basis for its further clinical application. METHODS: Seventy-two fracture models of Wistar rats were randomly divided into model control group, Shenggu Zijindan group and positive control group. After 2 weeks, 3 weeks and 4 weeks after modeling, localized osteophytes were made into paraffin sections. Immunohistochemistry technique was used to show TGF-β1 and bFGF for image quantitative analysis. RESULTS: During the fracture healing process, the positive expression of TGF-β1 and bFGF was observed in the callus. Gray-scale measurement results showed that the positive expression intensity of TGF-β1 and bFGF at the 2nd and 3rd week was found in the Zigu-zidan group and the positive control group. There was a highly significant difference from the model blank group (P<0.01). Conclusion: Shenggu Zijindan can promote the expression of TGF-β1 and bFGF and promote fracture healing.
