以斑马鱼(Danio rerio)雌性成鱼为研究对象,用不同剂量(0.1,1和10μg·L-1)的乙炔基雌二醇(17α-ethynylestradiol,EE2)对其进行暴露处理20d,并采用组织学方法对卵巢发育进行观察。组织学结果表明,不同剂量的EE2对斑马鱼卵巢发育均产生严重影响:卵巢和卵母细胞发育停止,减数分裂抑制,并出现卵黄积累期生殖细胞退化现象。为探讨上述影响可能的机制,采用半定量RT-PCR的方法检测了与斑马鱼卵子发育紧密相关基因vasa、scp3和igf3在EE2处理后的表达变化。研究结果表明,EE2处理之后,vasa基因的表达没有显著变化,而基因scp3、igf3的表达明显下调,且呈时间依赖性和剂量依赖性效应。因此研究推测EE2可能通过抑制igf3基因的表达和减数分裂抑制了斑马鱼卵子的发育和成熟。
The adults of Danio rerio were exposed to 17d-ethynylestradiol (EE2) with different dosages (0.1, 1 and 10μg · L-1) for 20 days, Histological methods were observed ovarian development. Histological results show that different doses of EE2 have a serious impact on the development of zebrafish ovary: ovarian and oocyte development stopped, meiosis inhibited, and yolk accumulation period germ cell degeneration. To explore possible mechanisms of the above effects, semi-quantitative RT-PCR was used to detect the expression changes of vasa, scp3 and igf3, which are closely related to zebrafish egg development after EE2 treatment. The results showed that there was no significant change in the expression of vasa gene after EE2 treatment, while the expressions of genes scp3 and igf3 were significantly down-regulated in a time-dependent and dose-dependent manner. Therefore, it is speculated that EE2 may inhibit the development and maturation of zebrafish eggs by inhibiting the expression of igf3 gene and meiosis.
