目的 探讨一氧化碳 (CO)对高血压大鼠主动脉重塑的影响。方法 32只Wistar大鼠随机分为四组 ,A组为正常对照组 ,B组为高血压组 ,C、D组为高血压干预组。测NO、CO、ET 1、AngⅡ浓度 ,计算主动脉中膜厚度 /管腔直径、中膜面积 /管腔面积的比值及血红素氧合酶 1蛋白表达强度。结果 B组大鼠血压高于A组 ,C、D组较B组降低 ,D组低于C组 (P <0 0 1)。血清NO浓度B、C、D组较A组降低 (P <0 0 0 1) ,D组较B、C组升高 (P <0 0 1) ;血浆CO浓度C、D组较A、B组升高 (P <0 0 0 1) ,D组高于C组 (P <0 0 0 1) ;血浆ET 1浓度B、C组较A、D组升高 (P <0 0 5 ) ;血浆AngⅡ浓度四组间无统计学差异。B组的主动脉中膜厚度 /管腔直径、中膜面积 /管腔面积的比值较A组升高 (P <0 0 0 1) ;C、D组较B组下降 (P <0 0 0 1) ;C、D组血红素氧合酶 1蛋白表达强于A、B组。结论 CO能防止高血压性主动脉重塑 ,其机制可能与CO降低血压、升高NO浓度及降低ET 1的浓度有关。
Objective To investigate the effects of carbon monoxide (CO) on aortic remodeling in hypertensive rats. Methods Thirty-two Wistar rats were randomly divided into four groups. Group A was normal control group, group B was hypertension group, group C and D were hypertension intervention group. The concentrations of NO, CO, ET 1 and Ang Ⅱ were measured. The aortic intima-media thickness / lumen diameter, intima-media area / lumen area ratio and heme oxygenase-1 protein expression intensity were calculated. Results The blood pressure of rats in group B was higher than that in group A, but lower in group C and D than in group B, and lower in group D than in group C (P <0.01). Serum NO concentrations in group B, C and D were lower than those in group A (P <0.01), group D was higher than group B and C (P <0.01) (P <0.01). The plasma ET 1 concentrations in group B and C were higher than those in group A and D (P <0 05). There was no significant difference among the four groups in plasma Ang Ⅱ concentration. In group B, the ratios of aortic media thickness / lumen diameter and medial area / lumen area were higher than those in group A (P <0.01); those in group C and D were lower than those in group B (P <0 0 0 1). The expression of heme oxygenase 1 in group C and group D was stronger than that in group A and group B. Conclusion CO can prevent the remodeling of hypertensive aorta. The mechanism may be related to CO decreasing blood pressure, increasing NO concentration and decreasing ET 1 concentration.
