目的应用H9c2心肌细胞系,观察雌二醇能否对缺氧状态下心肌细胞SERCA的活性及SERCA2a的表达产生影响。方法使用0,1nmol/L,100nmol/L浓度的雌二醇分别作用正常培养状态和缺氧培养的H9c2细胞5min,10min,20min、30min作为雌二醇对SERCA活性的短效作用研究。使用0.01nmol/L,1nmol/L,100nmol/L和1μmol/L的雌二醇分别作用缺氧培养条件下的去雌激素培养的H9c2细胞24h,作为雌二醇对SERCA活性和表达影响的长效作用研究。结果两种浓度的雌二醇不存在对SERCA活性的短效作用。缺氧能显著降低SERCA活性和表达(P<0.05),同时100nmol/L和1μmol/L的雌二醇能有效逆转上述作用(P<0.05)。结论 SERCA活性和表达的降低可能参与了缺氧状态下心肌细胞的病理生理改变,雌二醇可能对缺氧心肌细胞具有保护作用。
Objective To investigate whether estradiol can affect the activity of SERCA and the expression of SERCA2a in H9c2 cardiomyocytes. Methods Estradiol at concentrations of 0, 1 nmol / L and 100 nmol / L was used to study the short-term effect of estradiol on SERCA activity in normal and hypoxic culture H9c2 cells at 5 min, 10 min, 20 min and 30 min, respectively. The effect of oestradiol on SERCA activity and expression was studied by using oestradiol 0.01 nmol / L, 1 nmol / L, 100 nmol / L and 1 μmol / L, respectively, for 24 h in estrogen-depleted H9c2 cells under hypoxic conditions Effectiveness research. Results There was no short-acting effect on SERCA activity in both concentrations of estradiol. Hypoxia could significantly reduce SERCA activity and expression (P <0.05), while 100 nmol / L and 1 μmol / L estradiol could effectively reverse these effects (P <0.05). Conclusions The decrease of SERCA activity and expression may be involved in the pathophysiological changes of cardiomyocytes under hypoxic condition. Estradiol may have a protective effect on hypoxic cardiomyocytes.
