建立了培养周期短、再生效率高的麻疯树叶片再生体系.6-BA是诱导麻疯树叶片产生愈伤组织的关键外源激素,添加低浓度的2,4-D能增强6-BA的诱导效果.最佳愈伤组织诱导培养基是MS+2.0 mg/L 6-BA+0.05 mg/L 2,4-D,瘤状愈伤组织的诱导率高达91.98%±2.1%.光培养下产生的愈伤组织质地紧密,瘤状结构丰富,再生能力强.随着继代次数的增加,愈伤组织褐化加重,逐渐失去再生能力.不定芽再生培养基中,比较了3种细胞分裂素6-BA、KT和TDZ对诱导愈伤组织分化不定芽的影响.6-BA诱导不定芽的能力最优,其次是TDZ,KT最差.KT与BA的组合诱导不定芽再生的能力最强,最适的不定芽再生培养基为MS+0.5 mg/L 6-BA+0.5 mg/L KT+0.1 mg/L IBA+0.1 mg/L GA3,再生率mg/L)中均能产生根系,平均根系5.2个.对麻疯树叶片再生的各阶段进行了石蜡切片,揭示了不定芽的起源及发育的全过程,明确了不定芽起源于叶片的表皮细胞层.
