目的:探讨骨形成蛋白-2(BMP-2)在卵巢癌上皮性癌(EOC)中的表达检测及对卵巢癌细胞增殖影响的研究。方法:选择该院2007年4月~2010年7月收治的98例卵巢癌患者,提取患者肿瘤细胞后,采用SABC免疫组化方法和RT-PCR技术,检测卵巢上皮性肿瘤和正常卵巢组织中BMP-2蛋白及其mRNA的表达,分析其与卵巢癌细胞增殖及癌细胞组织分级之间的关系。结果:BMP-2蛋白在卵巢癌细胞组织中的阳性表达率为79.59%(78/98),与正常卵巢组织中的阳性表达率30.43%(7/23)比较有统计学差异(P<0.01);BMP-2蛋白与mRNA在卵巢癌组织高级别组(Ⅲ级、Ⅳ级)中的表达显著性高于低级别组(Ⅰ级,Ⅱ级),表明了随着癌细胞级别的升高,BMP-2的表达也逐渐增强。结论:BMP-2的表达对卵巢上皮性癌细胞增殖具有抑制的作用,同时BMP-2蛋白及其mRNA的异常表达可能与卵巢癌的恶性程度相关,因此可以作为检测肿瘤的一个标志物在临床中应用。
Objective: To investigate the expression of bone morphogenetic protein-2 (BMP-2) in epithelial ovarian cancer (EOC) and its effect on the proliferation of ovarian cancer cells. Methods: A total of 98 patients with ovarian cancer who were treated in our hospital from April 2007 to July 2010 were enrolled in this study. SABC immunohistochemistry and RT-PCR were used to detect the tumor cells in ovarian epithelial tumors and normal ovarian tissues BMP-2 protein and its mRNA expression, analysis of its relationship with ovarian cancer cell proliferation and cancer cell grade. Results: The positive rate of BMP-2 protein in ovarian cancer tissues was 79.59% (78/98), which was significantly different from that in normal ovarian tissues (30.43%, 7/23) (P <0.01) ); The expression of BMP-2 protein and mRNA in ovarian cancer high grade group (Ⅲ grade, Ⅳ grade) was significantly higher than the low grade group (Ⅰ grade, Ⅱ grade), indicating that as the level of cancer cells increased , BMP-2 expression also gradually increased. CONCLUSION: The expression of BMP-2 has an inhibitory effect on the proliferation of ovarian epithelial cancer cells. Meanwhile, the abnormal expression of BMP-2 protein and its mRNA may be related to the malignancy of ovarian cancer, so it can be used as a marker for the detection of tumors in clinical In the application.
