目的:建立同时测定血浆中茶碱、苯巴比妥和卡马西平浓度的方法。方法:采用RP-HPLC,色谱柱:ZORBAX SB-C18 (4.6 mm×250 mm,5 μm),柱温:30℃,内标:乙酰苯胺,流动相:甲醇-水(52:48),流速:1 ml·min-1,检测波长:254 nm。结果:茶碱、苯巴比妥和卡马西平的线性范围分别为:2.6-41.6 μg·ml-1(r=0.999 1),4.97-79.52 μg·ml-1(r=0.999 2), 1.31-20.96 μg·ml-1(r=0.999 7)。平均回收率分别为99.3%,99.4%,99.9%;日内及日间RSD均小于5%。结论:该法简单快捷,灵敏度高,结果准确,避免联合用药的影响,适用于临床血药浓度监测。
Objective: To establish a method for the simultaneous determination of theophylline, phenobarbital and carbamazepine in plasma. Methods: RP-HPLC was used. The chromatographic column was ZORBAX SB-C18 (4.6 mm × 250 mm, 5 μm). The column temperature was 30 ℃ and the internal standard was acetanilide. The mobile phase was methanol- , Flow rate: 1 ml · min-1, detection wavelength: 254 nm. RESULTS: The linear ranges of theophylline, phenobarbital and carbamazepine were 2.6-41.6 μg · ml-1 (r = 0.999 1), 4.97-79.52 μg · ml -1 (r = 0.999 2), 1.31-20.96 μg · ml-1 (r = 0.999 7). The average recoveries were 99.3%, 99.4% and 99.9%, respectively. The daily and intraday RSDs were less than 5%. Conclusion: The method is simple, rapid, sensitive, accurate and avoids the influence of combination therapy and is suitable for the monitoring of clinical plasma concentration.
