目的利用转录组技术获得非模式生物黑胸大蠊的大量转录组数据,为今后开展分子生物学研究打下基础。方法利用Illumina测序技术对黑胸大蠊Periplaneta fuliginosa进行转录组测序。测序结果取阈值e≤10~(-10)在SwissProt、NR、KEGG、COG、GO和Pfam等公共序列数据库进行比对。结果获得黑胸大蠊转录组信息数据量13.63 G,39576个unigene,总长度32 297 597 bp,最长27 693 bp,平均长度816 bp。共获得61 167个注释结果。发现黑胸大蠊单核苷酸的多态性位点(SNP)置换33 955个,颠换16 561个,找到2 666个微卫星序列。结论数量巨大的转录组信息为黑胸大蠊新基因的克隆和基因组学研究提供了有价值的信息。
OBJECTIVE: To obtain a large number of transcriptome data of non-model organism, Blattella patens, by using transcriptome technology, and to lay the foundation for molecular biology research in the future. Methods Illumina sequencing was used to sequence the transcriptome of Periplaneta fuliginosa. The results of the sequencing were compared with the public sequence databases of SwissProt, NR, KEGG, COG, GO and Pfam using the threshold e≤10 ~ (-10). Results The informative data of Blattella patens transcriptome was 13.63 G and 39576 unigene with the total length of 32 297 597 bp and the longest of 27 693 bp with an average length of 816 bp. A total of 61 167 annotated results were obtained. We found that 33 955 SNPs were replaced by 16 561 SNPs and 2 666 microsatellite loci were found. Conclusion A large number of transcriptome information provided valuable information for the cloning and genomics study of new genes in the cockroach.
