目的初步探讨心脉隆注射液对新生SD大鼠海马神经干细胞增殖分化作用的影响。方法分离新生SD大鼠海马组织,采用无血清及单克隆培养方式获得大量神经干细胞,将不同浓度心脉隆注射液加入第3代神经干细胞单细胞悬液中有血清贴壁培养并分实验组和对照组,应用免疫荧光及细胞化学染色检测Nestin、5-溴脱氧尿嘧啶核苷(BrdU)、微管相关蛋白2(MAP-2)、胶质纤维酸性蛋白(GFAP)的表达,计算MAP-2阳性细胞百分率。四甲基偶氮唑盐(MTT)比色法观察心脉隆注射液对神经干细胞增殖能力的影响。结果海马NSCs在分化后,各心脉隆注射液干预组MAP-2阳性细胞比例均较空白对照组增高(P<0.05),MTT结果显示培养5 d后各药物组NSCs增殖能力均较对照组增强(P<0.05)。结论心脉隆注射液具有促进神经干细胞增殖并向神经元方向分化的作用。
Objective To investigate the effect of Xinmailong Injection on the proliferation and differentiation of neural stem cells in neonatal SD rats. Methods The hippocampal tissue of neonatal SD rats was isolated, and a large number of neural stem cells were obtained by serum-free and monoclonal culture. Different concentrations of Xinmailong injection were added to the third generation of neural stem cells single cell suspension with serum adherent culture and divided into experimental group And control group, the expression of Nestin, BrdU, MAP-2 and GFAP were detected by immunofluorescence and cytochemical staining, and MAP -2 positive cell percentage. Effect of Xinmailong Injection on the Proliferation of Neural Stem Cells Observe with MTT Colorimetry. Results After differentiation of NSCs in hippocampus, the percentage of MAP-2 positive cells in each intervention group was significantly higher than that in control group (P <0.05). The results of MTT assay showed that the proliferation of NSCs in each drug group was higher than that in control group Enhanced (P <0.05). Conclusion Xinmailong injection can promote the proliferation of neural stem cells and differentiate into neurons.
